Middlesex, who have won the last five Championship games between the two counties, have named seamers Dan Evans and Gareth Berg in their 12-man squad.
The duo keep their places after taking seven wickets between them in Sunday's FP Trophy victory over Surrey.
Glamorgan open their Championship campaign looking to end a 54-year run without a first-class win at Lord's.
Paceman Jason Gillespie, all-rounder Jamie Dalrymple and opener Matthew Wood all make their debuts for the county.
Glamorgan boss Matthew Maynard is hoping for a good start to the campaign and an end to a poor sequence of Championship results at the London venue.
"We all know that if you have a good start to the season it can determine how that season goes," he said.
"The match at Lord's is a big game for us because we know one win can make such a difference.
"Historically, Lord's has always been a difficult venue for us to go and play, but we are a side who wants to change history and what better place to do it than at Lord's.
"The emphasis of our pre-season work has been to build up to this contest at Lord's so that we hit the ground running."
